Enzyme immunoassay for the quantitative determination of human ECP (eosinophilic cationic protein) in serum.

The ECP test kit is a sandwich ELISA and detects human ECP with a detection limit of 0.4 ng/ml. Cross-reactivity with EDN has not been observed. The solid phase is a microtiter plate well to which an anti-human ECP antibody (monoclonal) is bound. In the first step, patient or calibration serum is pipetted into the microtiter plate well. The ECP binds to the solid phase-bound anti-human ECP. After a washing step, an anti-human ECP antibody (polyclonal) labeled with horseradish peroxidase is pipetted into the microtiter plate well. An anti-human ECP/ECP/anti-human ECP (enzyme) complex is formed.


After a washing step, the substrate solution is pipetted into the microtiter plate. The enzymatic activity creates a colored solution from the substrate. The enzymatic reaction is then stopped by adding a stop solution. The amount of labeled anti-human ECP antibody bound is proportional to the amount of ECP released. The measurement is carried out in the photometer at 450 nm. A calibration curve is created from the measured calibration sera. The patient values are calculated using the calibration curve.
